Yes, it is specifically observed in mosques. This has always been the practice we received from Sunnah.

Then how we can say that prophet puh was doing Iteekaf while first revelation Occurred … and how it possible for him to do itekaaf in غار حراء

But he was doing iteekaf there, as in Sira ibn Hesham says and sir ghamidi also quotes that,
So on that mountain, where he was doing itekaaf ? Since you said that itekaaf is applicable in mosque not any other place

When you say sunnah Eiktikaf it is held only in the mosque. The prophet’s actions before the prophethood are not Hujjah. However, if one likes to sit in isolation or meditation, one may do that but that is not a sunnah aiktikaf. The prophet never did it outside the mosque. His wives did not do that in their houses adjacent to the mosque. They had to come to the mosques for Etikaf.
